Subculture Routine

Doubling time approximately 30 hours. Cells grow singly in suspension. Maintain cultures between 5-20 x 10⁵ cells/ml; 5% CO₂; 37°C. Thaw ampoule and perform a cell count, spin out the DMSO and resuspend in an appropriate volume of media to give a cell concentration of 5 x 10⁵ cells/ml. At subculture perform a cell count and maintain cultures between 5-20 x 10⁵ cells/ml, this tends to equate to a 1:3 split every 2-3 days.
