|Subculture Routine:||Split sub-confluent cultures (70-80%) 1:2 to 1:4 using 0.25% trypsin/EDTA; 7.5% CO2; 37°C. Suggested seeding density 2-4x10,000 cells/cm² when splitting. Seed at approx 6 x 10,000 cells/cm² when resuscitating from frozen. Following resuscitation or subculture the cells may take at least 48 hours to re-attach. They should be left without disturbance during this time to facilitate adhesion. These cells may take up to one week from resuscitation to establish and will require media changes during this time. The cells grow in discrete islands and do not form a monolayer within the flask. Cell counts should be performed at each subculture to monitor cell proliferation.|
