The fibroblast-like cell line named Tufted Deer has been derived from the lung of a male deer. Important Note - Species Query for the cell line ‘Tufted Deer': ECACC uses DNA barcoding to verify the species of cell lines. Using the Basic Local Alignment Search Tool (BLAST) DNA sequence alignment program the cytochrome C oxidase subunit I (cox1) gene sequence for the Tufted Deer (Elaphodus cephalophus) cell line has a match of 92% with Père David's Deer (Elaphurus davidianus) and a match of 88% with Tufted Deer. To assign a species the Quality Control criteria at ECACC requires a DNA barcode match of 95% or higher. ECACC has discussed this result with colleagues at the National Center for Biotechnology Information (NCBI) who host BLAST and Barcode of Life Database (BOLD) Systems. The conclusion is that the cell line named Tufted Deer is Deer but not Tufted Deer as originally described when deposited with ECACC. The exact species of Deer is not known. It should be noted however, that search engines such as BLAST allow open access and the sequences placed into them are not reviewed. This means the accuracy of the data in the databases the Tufted Deer sequence has been compared to cannot be guaranteed.

Split sub-confluent cultures (70-80%) 1:3 i.e. seeding at 2-4x10,000 cells/cm² using 0.05% trypsin or trypsin/EDTA; 5% CO₂; 37°C Culture attachment appears to be surface dependent, ECACC recommends the use of Corning Plastics.
