Cell Line Description

tsA201 is a transformed human kidney (293, ECACC catalogue no. 85120602) cell line stably expressing an SV40 temperature-sensitive T antigen. The cell line has been used in a variety of functional expression assays and has been reported to produce high levels of recombinant proteins. Transfection efficiency decreases over a period of several months, therefore it is advisable to lay down a frozen stock as a reserve. The cells have been eradicated from mycoplasma at ECACC. The identity of tsA201 and 293 has been confirmed by STR profiling.

Subculture Routine

Split sub-confluent cultures (70-80%) 1:2 to 1:8;i.e. seeding at 2-4x10,000 cells/cm² using ; mechanical agitation or EDTA solution; 5% CO₂; 37°C. Do not let cells become over confluent as this can affect morphology.

Culture Medium

DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
