Pluripotent mouse embryonic stem cell line. ES-E14TG2a cells are deficient in in HPRT and are resistant to 0.06mM 6-thioguanine.

Culture Conditions

Cell Type

Adherent monolayer of spheroidal cells on feeder layer of mouse primary embryonic fibroblasts or STO cells ECACC Catalogue 86032003 (irradiated or treated with mitomycin C to prevent cell division. The cells spontaneously differentiate and form embryonal structures in the absence of a feeder layer.

Culture Medium

DMEM + 2mM Glutamine + 1% NEAA + 0.1mM 2-Mercaptoethanol + 1000U/ml mouse inhibitory factor (LIF) + 1.5% sodium bicarbonate + 10% Foetal Bovine Serum. Renewal of medium every 2-3 days is required.
