Cell Line Description

The cell line TRA-171 has been established from freshly hatched larvae of Toxorhynchites amboinensis. After eggs had been surface-sterilised, they were allowed to hatch in MM / VP12 medium and were mechanically damaged at the anterior or posterior ends of the body in 0.05% trypsin. After washing and cultivation at 22 - 25°C small patches of cells were visible leading to the isolation of the cell line TRA-171 after 72 days in culture. The cells have been adapted to grow in a 1:1 mixture of Leibovitz L-15 and Mitsuhashi and Maramorosch media plus supplements. Four serotypes of laboratory adapted and three type strains (1, 2 and 3) of un-adapted dengue virus will replicate in the cells at 28°C.

Unless specified otherwise, at ECACC we routinely handle all of our cell lines at containment level 2 in accordance with the ACDP guidelines (Advisory Committee on Dangerous Pathogens) (UK). All cell cultures have the potential to carry as yet unidentified adventitious agents. It is the responsibility of the end user to ensure that their facilities comply with biosafety regulations for their own country.
