Cell Line Description

HeLa DH cells are the wildtype that has been used for connexin-transfection assays and subsequent analysis of permeability and gap junction formation. The wildtype exhibits stable membrane potentials, very low gap junctional conductance and high stability under double whole-cell patch-clamp measurements. Gap junction plaques were not detected but using antibody directed against connexin 45 identified gap junction channels as dispersed particles in non typical gap junction areas.

Split sub-confluent cultures (70-80%) 1:3 to 1:8 i.e. seeding at 1-3x10,000 cell/cm² using 0.05% trypsin or trypsin/EDTA; 5% CO₂; 37°C.
