Cell Line Description

CAD cells (Cath.-a-differentiated) are a variant of a CNS catecholaminergic cell line established from a brain tumour in a transgenic mouse carrying wild-type SV40 T antigen under transcriptional control of rat tyrosine hydroxylase promoter. CAD has lost the immortalising oncogene and expresses neurone-specific proteins and synaptic vesicle proteins. CAD will differentiate to form neuronal processes in serum free conditions.

Subculture Routine

These cells may take 1-2 days to attach and will initially grow in loosely attached small islands. The islands will increase in size in the following 1-2 days and should be sub-cultured when the flask reaches 70-80% confluence. Cells should be removed by gentle knocking of the flask and pipetting. Split cultures 1:4 to 1:10 seeding at 3 X 10⁴ cells/cm² ;5% CO₂; 37°C.
