Skeletal muscle phenotype, no cardiac properties despite origin. Myotubes formed at confluency respond to acetylcholine. Differentiation improved by reducing serum concentration to 1%.

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:10 i.e. seeding 1-3x10,000 cells/cm² using 0.05% trypsin or trypsin/EDTA; 5% CO₂; 37°C. Important to passage at sub-confluence, reclone periodically to keep differentiated characteristics. Cells will fuse to form myotubes if allowed to reach confluency.

DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
