Subculture Routine

Split sub-confluent cultures (70-80%) 1:5 to 1:10 i.e. seeding at 5x1,000 to 1x10,000 cells/cm² using 0.1% cold PBS and 0.05% cold trypsin. Place flasks at 4°C for 5-10 min. or until cells detach; 5% CO₂; 37°C. Pre-treat culture flasks with porcine gelatin (Sigma G-2625) overnight at room temperature, or for at least 2hr at 37°C. Serum is only used to initially resuscitate from frozen and should be added (1%) prior to cryopreservation.

EMEM (EBSS) + Ham's F12 (1:1) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 0.1% BSA.
