Cell Line Description

DMS 92 was obtained from a bone marrow aspirate of an untreated 39 year old male at time of diagnosis. The isolated cells were identified as small cell carcinoma of the lung (SCLC). DMS 92 cells secrete adrenocorticotropin (ACTH), bombesin (BN), oxytocin-neurophysin (OT-NP) and 17 beta-estradiol. Cells produce tumours in athymic nude mice. The Y chromosome could not be detected in this cell line by short tandem repeat (STR)-PCR analysis when tested at ECACC. It is a known phenomenon that due to the increased genetic instability of cancer cell lines the Y chromosome can be rearranged or lost resulting in lack of detection. The cell line is identical to the source provided by the depositor based on the STR-PCR analysis.

Culture Conditions

Subculture Routine

On resuscitation, a centrifugation step to remove the cryoprotectant is essential. Rapidly thaw the frozen ampoule in a water bath at 37°C for 1-2 minutes. Transfer the contents to a centrifuge tube and slowly add 5-10ml of pre-warmed growth media. Remove a sample for counting. Centrifuge at 100 x g for 2-3 minutes to pellet cells and seed at approximately 5-7 x 105 cells/ml.

Cell line is slow to proliferate and may take up to 20 days to establish. Once established maintain cultures between 3-9 x 105 cells/ml.

Recommended growth conditions: 5% CO2, 37°C.

Waymouth's MB 752/1 + 2mM Glutamine + 10% Foetal Bovine Serum (FBS) (Heat Inactivated).
