Cell Line Description

EMT6/VCR/R is a drug-resistant variant of the mouse mammary tumour cell line EMT6/P (ECACC catalogue no. 96042344). It was derived from the parent line by continuous growth in increasing concentrations of vincristine. EMT6/VCR/R was reported to be cross-resistant to other drugs of the multidrug-resistance (MDR) phenotype, showed reduced intracellular drug accumulation and hyperexpression of P-glycoprotein.

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:6 using 0.05% trypsin or trypsin/EDTA; 5% CO₂; 37ºC. It is recommended to resusitate cells in drug-free medium. Add drug after growth is established.

Culture Medium

MEM(EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 1µg/ml vincristine + 10% Foetal Bovine Serum (FBS).
